Kronik inflamasyon indikatörü olan IL-10 prostat kanser gelişiminde etkin olabilir. Bu çalışmada IL-10 ekspresyonunu etkileyen promotor bölge polimorfizmleri ile prostat kanser gelişimi arasındaki ilişkinin ortaya konması hedeflenmiştir. Çalışmada benign ve malign prostat tümörlü hastalardan alınan doku örneklerinden DNA izolasyonu yapılarak, elde edilen DNA’larda, IL-10-1082 ve IL-10-592 polimorfizmleri Real Time PCR ile çalışılmıştır. Sonuç olarak malign prostat kanserli olgularda IL-10-1082 GG (wildtype) genotipi görülme sıklığı mutant AA ve heterozigoz GA genotiplerine göre daha fazla bulunmuştur. It has been known that cronic intraprostatic inflammation may have a role in prostate cancer pathogenesis. The relationship between prostat cancer patogenesis and IL-10, which is cronical inflamation indicator, is not clear. This study may contribute to understand the relationship between promoter polymorphisms which effects the IL-10 expression and prostate cancer diagnosis and prognosis. During the study, DNA isolated from benign and malign prostate tissue samples. Real Time Polymerase chain reaction performed using specific IL-10 primers to detect possible polymorphisms in isolated DNAs. In conclusion we have demostrated that IL-10-1082 GG (wildtype) gene polymorphism is more prevalent in malign PCa patients compared GA and AA genotypes. We also observed that the mutant heterozygot IL-10-592 CA polymorphisms is positively related to increased PSA levels. | URI: | https://hdl.handle.net/11499/365 |
