Behçet hastalarında HLA-B51 ekspresyon düzeyleri üzerine çalışmalar

Abstract

Behçet Hastalığı, etiolojisi tam olarak açıklanamayan tekrarlayan kronik multisistemik bir hastalıktır. Behçet hastalığında genetik ve çevresel etkenlerin katkıları araştırılmıştır. Bununla birlikte Behçet hastalığı ve HLA sınıf I grubu içinde yer alan HLA-B51 molekülü arasındaki güçlü bir ilişkinin bulunması hastalığın gelişiminde genetik faktörlerin işe karıştığını açıkça göstermiştir. Bu tez çalışmasında, uluslararası standartlara göre, Behçet hastalığı tanısı alan bireylerde HLA-B51 molekülünün frekansı ile bu genin ekspresyon frekansının saptanması amaçlanmıştır. 50 normal sağlıklı birey ile 35 Behçet hastalığı tanısı almış kişiden alman kan Örnekleri araştırmamızda kullanıldı. Kan örneklerinden DNA ve RNA izolasyonu yapıldı. HLA tiplerinin ekspresyonlanının tanımlanması için RT-PCR tekniği kullanıldı. HLA-B5I genini çoğaltabilecek primerler ile bölgeye özgün prob laboratuvarımızda tasarlanmıştır. HLA-B51 bölgesi için hazırlanan primerler kullanılarak yapılan DNA-PCR yöntemi ile normal sağlıklı bireylerde %58 oranında, Behçet hastalarında %63 oranında bölge çoğaltımı elde edilmiştir. RT-PCR yöntemi ile 29 normal sağlıklı bireyde %90 oranında, 22 Behçet hastasında %72 oranında bölge çoğaltımı elde edilmiştir. Sonuç olarak, Behçet hastaları ile sağlıklı normal gruplardaki HLA-B51 ekspresyonunda önemli bir farklılık olmadığı kanısına varılmıştır. 46

Behçet' s Disease is a chronic multisistemic disease ethiopathogenesis of which is not well known. The development of the disease is considered in light, environmental and genetic predisposition when the geographical locality and incidence are concerned. However; the exact relationships between HLA-B51 molecule and the development of Behcet's Disease is related with genetic factors. In this thesis, we aimed an investigation of the HLA-B51 frequency and its expression level, in individuals who have been diagnosed as Behçet' s Disease patients according to international standards and in normal subjects. For the investigation of these data, we used 35 Behçet' s Disease patient and 50 normal healthy controls. Blood samples were taken from these individuals for the isolation DNA and RNA. The RT-PCR technique was used for the identification of expressed HLA types. The specific primers and prob were designed in our laboratory. For the HLA-B51 region specific PCR amplification, we observed that the presence of HLA-B51 was 58 % in controls, and 63 % in Behçet' s Disease patients. The expression patterns obtained by RT-PCR technique showed 90 % expression in controls, and 72 % in Behçet' s Disease patients. As a result; we concluded that there is no relationship in between Behçet' s Disease and the presence and expression of HLA-B51 compared to normal healty controls. 47
