Publisher: | Nova Science Publishers, Inc. | Abstract: | Stereological methods provide precise and efficient image analysis opportunities. However, in order to obtain three-dimensional information from two-dimensional images, a larger number of microscopic views are needed compared with semi-quantitative methods. This disadvantage can adversely affect the motivation of image analysis students. Image analysis programmes coupled with top-of-the-range computers are useful for overcoming that disadvantage. However, not all laboratories possess such systems due to their high costs. The present study discusses a system that includes a simple cameraattached laboratory microscope and a stereology image analysis software (SHTEREOM) capable of provide quantitative stereology estimations. SHTEREOM has been designed to be adaptable to any computer with Windows software. SHTEREOM version 1.5 possesses two modules. These are used for volume estimations and object number estimations by means of the optical fractionator method. The software does not need any specific hardware. It therefore requires only need a computer with screen card, which can transfer the image captured by the camera mounted light microscope. In addition, the software needs to be calibrated before the estimations commence. In order to perform volume estimations, the thickness of the sections which can be measured by the methods described in the literature should be entered into the software. Analyses can be carried out on photographs of sections taken in line with stereology rules and sampling strategies. Photographic images can be used for that purpose. However, each photograph should have a scale bar, and the section thickness should be known. The coefficient of error is automatically calculated, and section and total particle numbers are transferred to an Excel spread sheet. SHTEREOM is software that can be used by all researchers due to is affordability and ease of use. © 2024 Nova Science Publishers, Inc.
