Abstract: | Kokoreç, ülkemizde sık tüketilen sakatatlardan biridir. Literatür incelendiğinde kokoreçte biyoaktif peptitlerin analizi ve aminoasit profillerinin incelenmesi konusunda çalışmaya rastlanmadığı için kokoreçte biyoaktif peptitlerin incelenmesi araştırma konusu olarak tercih edilmiştir. Çalışmada temizlenmiş ve 1. pişirme işlemleri tamamlanmış kokoreçten biyoaktif peptitler izole edilmiş bunların biyoaktiviteleri incelenmiştir. Bu amaçla kokoreçten elde edilen ekstraktlar, hazırlanan solüsyonla beraber HPLC cihazından geçirilerek fraksiyonları toplanmıştır. Toplanan peptit ve protein fragmentleri, HPLC-DAD cihazı ile aminoasit analizi yapılarak içerdiği aminoasitlerine ayrılan bileşen konsantrasyonuyla bağlantılı olarak belirlenmiştir. Bu sonuçlara dayanarak ANOVA analizi yapılmış ve sonuçlar özetleme ve grafiklendirme ile istatistiksel açıdan incelenmiştir. Toplanan peptit ve protein fragmentleri LC/MS cihazında incelenerek peptit dizilimleri elde edilmiştir. Bu dizilimler BIOPEP veri tabanıyla incelenerek biyoaktif peptit aktiviteleri tespit edilmiştir. Ayrıca kokorecin antioksidan aktivitesinin incelenmesi spektrofotometre ile DPPH analizi ile hesaplanarak kokoreç peptitlerinin antioksidan aktiviteye sahip olduğu tespit edilmiştir. Kokorec is one of the most commonly consumed offal inTurkiye. Since there are no studies on the analysis of bioactive peptides and amino acid profiles in kokorec in the literatüre. The aim of the study was to investigate the bioactivity of the cleaned and pre cooked kokorec samples in vitro and to investigate the kokorec sample from different parts with amino acid analysis and antioxidant activities. For this reason, the extracts obtained from kokorec were passed through the HPLC device with the prepared solution and the fractions were collected. The aim here is to separate the sample into its compounds and analyse the peaks for each compound. Amino acid analysis is an important approach for the characterisation of protein and peptide based products. In addition, the amino acid composition determined can provide a measure of sample purity by confirming sample identity. For this purpose, the collected peptide and protein fragments were determined in relation to the concentration of the component separated into its amino acids by Amino Acid Analysis with HPLC-DAD device. Based on these results, ANOVA analysis was performed and the results were statistically analyzed by summarizing and graphing. The results showed that there was a strong correlation between the amino acid species, samples and their abundance. The collected peptide and protein fragments were analyzed by LC/MS liquid chromatography and peptide sequences were obtained (Ege MATAL). These sequences were analyzed by BIOPEP database and bioactive peptide activities were determined. In addition, in order to examine the antioxidant activity of kokorec, it was determined that kokorec peptides have antioxidant activity with DPPH analysis. |
